Vash commented that some of the cultures she had encountered in the Gamma Quadrant had histories spanning millions of years. (DS9: "Q-Less") So what are they? How advanced are they? What do you think is out there? Why has the series been more active on exploring it?
The Vulcans were at the forefront of the exploration of the Quadrant, beginning only months after the wormhole was discovered...the Vulcans made many discoveries, including the remains of the Hur'q race. (DS9: "The Sword of Kahless"). What could others, including Starfleet, yet discover?
The IKS Toh'Kaht was among the first ships to explore the quadrant in an effort to establish colonies on the other side of the wormhole. They also operated freighters to the quadrant. (DS9: "Dramatis Personae", "The Storyteller"). What happened to the colonies? Were the colonies successful?
What about the Ferengi's attempts to make profit or the first Bajoran colonies?

I would definitely recommend regarding the DS9 re-launch novels especially the MISSION GAMMA mini-series.

several colonies and ships were destroyed by the jem'hadar (season 2; season finale: The Jem'hadar)
I'm hoping STO will take us to the Gamma Quadrant at some point....
